ABSTRACT

@#Objective - ( )- ( ) To observe the effects of renin angiotensin Ang aldosterone system RAAS in workers exposed to Methods - - occupational noise. Forty five workers with suspected occupational noise induced deafness were selected as noise , , exposure group using convenient sampling method. According to their tinnitus symptom noise exposure intensity and work age - , , they were divided into no tinnitus and tinnitus subgroups <90 dB and ≥90 dB subgroups work years <10 years and ≥10 years subgroups. Another 45 workers with no occupational noise exposure history were selected as control group. The levels of plasma ( ), , , renin activity PRA AngⅠ AngⅡ and aldosterone of the two groups were detected and the aldosterone to renin activity Results ratio was calculated. The diastolic blood pressure of the noise exposure group was higher than that of the control group [( )vs( ) ,P ] , 80±7 76±8 mmHg <0.05 . However there was no significant difference in systolic blood pressure between the two (P ) ( : groups >0.05 . The level of plasma AngⅡ in the noise exposure group was higher than that in the control group median vs ,P ) ( P ) 100.98 65.43 μg/L <0.05 . There was no statistical significance in other indexes between the two groups all >0.05 . The ( : plasma AngⅡ level in < 90 dB subgroup in the noise exposure group was higher than that of the control group median 123.16 vs ,P ) 65.43 μg/L <0.05 . There was no statistical significance in other indexes among the two subgroups of tinnitus symptom or ( P ) work age in the noise exposure group and the control group all >0.05 . There were no significant differences in the abnormal , ( P ) rates of PRA AngⅡ and aldosterone in plasma between the noise exposure group and the control group all >0.05 . Conclusion Occupational noise exposure may affect RAAS and lead to increased plasma AngⅡ levels in the workers. - Tinnitus and work age may not affect RAAS in occupational noise exposure workers.

ABSTRACT

Objective To investigate the effects of Bushen Jianpi Prescription on tumor growth in subcutaneous colorectal cancer xenografts in mice and expressions of VEGF and MMP-7; To discuss its mechanism of anti-colon cancer. Methods Nude mice were inoculated subcutaneously into human colon cancer cells to establish human colon cancer xenograft models. Nude mouse models of subcutaneous colorectal cancer xenografts were randomly divided into model group, 5-FU group, Bushen Jianpi low-, medium-, and high-dose groups. Each medication group was given relevant medicine for three weeks. Six tumor-bearing mice in each group were sacrificed. Tumor mass was measured, and the relative inhibition rate was calculated. The serum levels of VEGF and MMP-7 were measured by ELISA. The remaining tumor-bearing mice were used to observe the survival time. Results Compared with model group, the weight of tumor were reduced and the median survival time of mice were prolonged in Bushen Jianpi groups, as well as the expression levels of VEGF and MMP-7 significantly decreased Bushen Jianpi groups. Conclusion Bushen Jianpi Prescription can inhibit the growth of human colon cancer xenografts in nude mice and prolong the survival time of tumor-bearing mice, which may be related to the down-regulation of the expressions of VEGF and MMP-7.

ABSTRACT

<p><b>OBJECTIVE</b>To explore in vivo metabolic changes in abnormal savda patients with different types of tumor.</p><p><b>METHODS</b>A total of 142 abnormal savda patients with common cancer types were enrolled in this study, and 50 healthy volunteers were recruited as the control group. For each sample, the H Nuclear Magnetic Resonance (NMR) based metabonomic analysis was performed. The free attenuation signal was computed subsection integral. Data obtained were analyzed by the Orthogonal Partial Least-Squares Discriminant Analysis (OPLS-DA).</p><p><b>RESULTS</b>Compared with the control group, leucine, isoleucine, valine, histidine, phenylalanine, tyrosine, alanine, creatine, lactic acid, inositol, alpha-and beta-glucose, unsaturated lipids, very low density lipoprotein (VLDL) significantly decreased (P <0.05), while glycoprotein and carnitine significantly increased (P <0. 05) in the abnormal Savda group.</p><p><b>CONCLUSION</b>Abnormal savda patients with different types of tumor had similar metabonomics changes.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the effects and molecular mechanism of action of Changweiqing (CWQ) in reversing multidrug resistance by observing its impacts on nuclear translocation of Y-box binding protein-1 (YB-1), multi-drug resistance gene (MDR1) expression and P-glycoprotein (P-gp) expression in human colon cancer cell line HCT8/V with drug-resistance induced by vincristine.</p><p><b>METHODS</b>Cultured HCT8/V cells were exposed to the CWQ-containing rat serum prepared by drug perfusion. YB-1 expressions in cell plasma and nuclei were examined by Western blot; the binding activity of YB-1 to MDR1 gene promoter sequences was detected by electrophoretic mobility shift assay (EMSA); the mRNA transcription levels of MDR1, YB-1 and multi-resistance related protein (MRP) were examined by RT-PCR; the expression of P-gp on cell membrane was determined by flow cytometry. Results Along with the increasing drug's concentration of CWQ-containing serum from 1.25% up to 2.5% and 5%, the expressions of YB-1 decreased in HCT8/V cell nuclear and increased in cytoplasm gradually; the binding activity of YB-1 to MDR1 gene promoter weakened (P < 0.01), MDR1 mRNA expression and fluorescence intensity of P-gp on cell membrane attenuated (P < 0.05 or P < 0.01), while YB-1 and MRP mRNA unchanged (P > 0.05).</p><p><b>CONCLUSION</b>CWQ could reverse the drug-resistance of colon cancer cells by influencing nuclear translocation of YB-1 and reducing the expression of MDR1/P-gp.</p>
